Offer Description

The emergence of new infectious agents, caused by globalization, climate change, increasing of the population, etc. leads to the need of new drugs and must be considered in a one-Health approach since the barrier between species is limited. As an example, the magnitude of the morbidity and mortality during the Covid-19 pandemic showed that the discovery and the development of effective antiviral drugs are an urgent priority. Moreover, phenomena of microbial resistance lead us to consider new molecular structures and new therapeutic targets. 1 We propose to combine the needs of antivirals and antimicrobials by appropriate modifications of fusion viral peptides. 2

The objectives of this project are to study the antiviral and antibacterial properties of viral fusion peptides. After selection of relevant viral fusion peptide sequences, native hydrophobic sequences as well modified sequences will be synthesized to study their ability to inhibit the viral fusion step. Different modifications will be introduced in order to allow interaction with bacterial membranes to change physicochemical properties and improve their amphipathic character. 3 Membranotropic properties will be evaluated by spectrofluorescence. 4 Because antimicrobial peptides often act via a pore formation, we will then study the ability of the new analogues to form pores in the bacterial wall and their behavior inside the pores. 5 These properties will be correlated with their biological activities (antiviral: University of Gdansk, antimicrobial: CY University) and to conformational studies (University of Naples, University of Paris-Saclay).
