  • - preparation and exploitation of nanopore devices, in particular, protein nanopore - evaluation and modification of different protein nanopores - preparation and purification of GAG oligosaccharides - Complementary structural analyses for the structural study of oligosaccharide libraries (chromatography, electrophoresis, NMR, mass spectrometry, among others) - processing of nanopore data and their organization in a database - Exploit the analytical results for publications, - Participate in the writing of scientific articles. - Apply the rules of hygiene and safety of the field

    The post-doctoral researcher will carry out his/her work in the Analysis, Modeling, and Materials for Biology and Environment Laboratory (LAMBE, CNRS UMR 8587, Université Paris-Saclay, Evry Univ. site), with the mission of analyzing bioactive polysaccharides glycosaminoglycans (GAGs) at the single molecule scale using nanopore detection with the objective of providing the first analytical method for sequencing these polysaccharides. This work is part of a national research program (ANR) for the period 2021-2024 and will be carried out under the scientific responsibility of the thematic leader. The different axes of the research project include: - Implementation of the nanopore biophysical system and related data processing, - constitution of structurally defined libraries of GAG oligosaccharides - Implementation of enzymatic transformation processes of GAGs, - detection and structural characterization by nanopore of GAGs of biosynthetic origin or chemical synthesis of variable size, sequence, and organization, and comparison with other analytical methods (chromatography, electrophoresis, NMR, mass spectrometry, among others), - Establishment of databases of structural signatures.

    The LAMBE is a CNRS unit (UMR 8587) at the University of Paris-Saclay, Evry Univ. site, whose activities fall under the "Chemistry and Life" and "Physical, Theoretical and Analytical Chemistry" sections of the CNRS Institute of Chemistry (INC). Its research topics mainly concern the structure/reactivity of biomolecules, in particular in the field of glycosciences. To this end, the laboratory exploits and develops methods based among others on biomimetic nanopores from their biosynthesis, characterization, modification, and microfluidic integration to study the dynamics in a confined environment, and for single molecule analysis. The laboratory also has expertise in mass spectrometry (MS) applied to the study of biomolecules, and operates a platform of seven mass spectrometry instruments. All the equipment and expertise necessary for the realization of the project are available in the laboratory.

    Eligibility criteria

    The candidate must have a Ph.D. in a field related to biochemistry (glycosciences, protein chemistry), biophysics (single molecule), or bioanalytical chemistry (separation methods, chromatography, mass spectrometry, and coupled methods applied to biomolecules). Strong knowledge of biochemistry, biophysical methods, and analytical chemistry (preparation, purification & detection) is required. Knowledge of bio-structural methods is desirable. The candidate will be motivated to take on scientific challenges. The candidate will demonstrate good writing skills with a good command of written English, and the ability to publish and promote research work. The candidate will have the ability to work in a team and in a network of partners.
