Offer DescriptionThe aim is to identify key transcription factors involved in reprogramming lipid metabolism by temperature in the minimal model marine microalga Ostreococcus tauri.
Based on temperature-dependent differentially expressed genes and differential membrane proteomics, the first objective is to extract candidate TFs involved in early reprogramming of lipid metabolism. The first step is to identify candidate TFs by combining bioinformatics (generation of gene regulatory network and promoter analysis, in collaboration with an expert in computational biology) and biochemical (DNA affinity purification of TFs) approaches. The next step is to 1) validate the binding of TFs to promoters by yeast 1-hybrid, 2) identify, in vivo, the pan-genomic target genes of the selected TFs using chromatin immunoprecipitation sequencing (ChIP-Seq) using O. tauri transgenics for TFs fused to a tag. The second objective is to evaluate the physiological function of the selected candidate TFs, in particular for temperature-induced reprogramming of lipid metabolism. To achieve this objective, O. tauri mutants (overexpressors and knockdown/out) for the selected TFs will be analysed to test their ability to remodel their membrane lipids and acclimatise to temperature (detailed lipidomics).
