  • The CIML is a joint research unit of the CNRS, INSERM and Aix-Marseille University. It is composed of about 200 people and 16 research teams and is located in the Luminy Science and Technology Park in Marseille (France). Within the CIML, the student will work in the research team "Dendritic cells and antiviral immunity" directly under the supervision of the team leader, Marc Dalod. The work performed will be part of a collaborative ANR research project ("nRegSKIN") coordinated by Dr. Mei LI (IGBMC, Strasbourg) in which Marc Dalod's team is partner.

    Project title: Role in the homeostasis of the skin and in local antiviral defences of the cross-talk between plasmacytoid dendritic cells, other dendritic cell types and regulatory CD4 T lymphocytes.

    Missions: The student will study in vivo in mice the functional interactions in the skin and lymph nodes between plasmacytoid dendritic cells, other dendritic cell types, and regulatory CD4 T cells, and their impact on antiviral immunity, in a model of cutaneous herpes simplex virus type 1 (HSV-1) infection. The student will also determine whether, and how, the induction of atopic dermatitis in a mouse model could modulate this process in a manner impairing antiviral immunity. This could help explain the recurrent HSV-1 skin infections observed in a fraction of patients suffering from atopic dermatitis, sometimes causing herpetic eczema with potentially fatal complications. These studies will be performed by combining different tools and technologies, including new mutant mice specifically targeting certain types of dendritic cells, analysis of innate and adaptive immune responses ex vivo by flow cytometry and in vivo by confocal microscopy, global tissue responses by sequencing of RNA extracted from skin biopsies, transcriptomic responses of immune, epithelial or stromal cells by single cell or single nucleus RNA sequencing.

    Activities: 1) Breeding and PCR genotyping of different mouse lines required for the research project experiments. 2) Animal experimentation: handling of alert mice, including skin infection with HSV-1, after local scarification. 3) Isolation of skin and lymph node cells, antibody labelling and acquisition by multiparametric flow cytometry. 4) Harvesting of skin and draining lymph node, preparation of sections of these organs, antibody labelling and analysis by multiparametric confocal spectral microscopy. 5) RNA sequencing at the single cell or nucleus level, after isolation of cells or nuclei from the skin and draining lymph node.

    Skills: 1) Excellent level of knowledge of Immunology 2) Experience in care and use of mice for experimentation (practitioner/operator level wished) 3) Proven experience in molecular biology: DNA and RNA extraction, PCR, qPCR (strongly wished) 4) Management of mouse lines, genotyping 5) Very good knowledge of histology and microscopy techniques, preferably confocal, and/or of flow cytometry (strongly wished) 6) Good knowledge of the English language, presentation in English of own results during team meetings
